UV 기술, 제약·음식료품 분야 적용 ‘적합’

화학물질 미사용·살균 및 소독 효율성 우수
설치 용이·시설 운영 부담 최소화…정비·보수 불필요

제약 및 음식료품 산업의 제품은 직접적으로 가공되지 않은 상태로 소모되거나 활성적으로 소비되는 제품으로 변형되어 사용되는 등 다른 산업 분야와는 다른 독특한 특성을 갖고 있다. 점차 규제가 강화되고 안전성이 중요시되는 글로벌 시장의 형성으로 이러한 산업은 수질 및 미생물에 대한 안전(biosecurity)을 엄격히 준수해야 한다.  이에 대해 Hanovia사의 기술이사인 Mark Aston은 UV(Ultra Violet) 기술이 효율성면에서 케미컬을 사용하지 않는 특성 등의 이유로 음식료품 산업 분야 적용에 적합하다고 주장하고 있다. 그 내용을 번역했다.

 

제조 공정에서 사용되는 물은 독성물질, 미생물 유해물질(biohazards), 불필요한 유기성 물질 등을 제거하는 것이 필수적이며, 이 과정에서 맛, 냄새 및 반응생성 물질 등 부산물이 발생되지 않아야 한다. 제조과정의 초기공정에 사용되는 물은 먹는 물 수준의 원수가 사용되기도 하나, 미생물 영향에서 완전하게 자유롭지 않은 물이 사용되어 물 속에 포함되어 있는 미생물의 활성화 작용으로 일부 유해한 수준으로 성장이 이루어지기도 한다.

그러나 UV(Ultra Violet) 기술은 살균 및 소독에서 효율성이 뛰어나며 케미컬을 사용하지 않는다. 이러한 적용에 매우 적합한 기술로써 박테리아, 바이러스, 효모균 (yeasts), 사상균 (moulds) 등 잘 알려져 있는 모든 종류의 병원성 미생물(pathogenic), 그리고 부패성 미생물(spoilage microorganism)을 제거하는 기능을 갖는다. 또한, UV 기술 설비의 사용에 특별한 정비·보수의 업무가 필요하지 않으며 환경 친화적인 기술로 알려져 있다.

It is vital to treat the water used in the manufacturing process to remove toxins, biohazards and any unwanted organic materials, while at the same time not incurring any residual taste, colour or reagents that could affect another part of the downstream process. In these industries the ‘raw’ potable water they source at the start of the process may not be completely free of bio-challenges, as needed for their individual processes, resulting in active growth of microorganisms to harmful levels.
UV(Ultra Violet) is ideal for these applications as it is both effective and chemical-free. It eliminates all known pathogenic and spoilage microorganisms, including bacteria, viruses, yeasts and moulds (and their spores) and is low maintenance and environmentally friendly.

미생물 안전의 중요성(The importance of biosecurity) 

미생물 안전(biosecurity)에 대한 중요성은 우선 공급자와 사용자 관점에서 충분한 이해가 있어야 한다. 즉, 공급자 측에서는 적절한 용량크기의 시스템이 제공되어야 하며 사용자 측에서는 비용투자의 경제성을 보장해주는 양자간 적절한 균형 관계가 수립되어야 한다.

UV 기술의 시스템이 특정 박테리아, 바이러스 및 미생물 유해물질의 제거를 목표로 하는 경우, 이러한 유해물질의 실제 감소에 필요한 UV 조사의 정확한 주입량이 최소가 되도록 설계·제작되어야 하며 챔버 내 이동 과정 중에 적절한 모니터링이 되어야 한다.

이와 같은 목적으로 다음의 3가지 형태의 시스템이 주로 사용된다.
- 최소한의 delivery dose를 보장하는 최소의 램프강도를 갖는 시스템
- 성능이 이미 알려져 있으며 boisecurity 보증을 제공하고 있는 시스템
- delivery를 고려한 정확한 dose를 모니터링하며 시스템 및 지속적으로 deliver 컨트롤 가능한 시스템 파워를 모니터링하는 기능의 성능보증이 이루어진 시스템

Biosecurity is very important to understand from both the supplier and customer perspective. It is a fine balancing act between providing an appropriately-sized treatment system on the one hand while ensuring viable cost of ownership on the other.
Where a UV system is required to target a specific bacteria, virus or other bio-hazard, the precise minimum dose needed to achieve the required reduction of that hazard must be well-characterized and monitored in delivery.
Three general types of system can be used for this task:
- Very cost-effective systems monitored for a minimum lamp intensity that achieve a minimum delivered dose which is not defined
- Cost-effective systems that have been referenced to known validations and UV disinfects water without chemicals or pasteurisation. therefore offer an assured biosecurity but without the absolute cost of a full validation
- Costly systems that have been fully validated to monitor the precise dose delivered and the power of the system controlled to continually deliver that required dose.

UV 살균기술의 원리(How UV disinfection work) 

UV광선은 가시광선과 X-ray 광선사이에 분포하는 전자계 스펙트럼의 일부로 UV광선 스펙트럼은 100∼290㎚사이에 분포한다. 특히 265㎚의 peak effective 파장에서 강력한 살균능력(germicidal) 효과를 갖고 있다. 이 파장에서 UV광선은 멤브레인 셀 내부에 침투되어 DNA를 파괴, 결과적으로 재생이 되지 못할 정도로 파괴시키는 역할을 함으로써 미생물들을 효과적으로 제거하게 된다.

전형적인 UV 시스템은 보호용 석영관(protective quartz sleeve) 내에 장착되는 UV 램프로 구성되며 이들은 원통형의 스테인리스강 챔버에 설치된다. 처리가 이루어지는 원수는 한쪽으로 흘러 들어가 챔버의 전체 길이를 따라 통과된 후 다른 쪽으로 배출된다. 실제적으로 자연상태의 원수, 필터처리된 공정수, 점성의 당류(viscous sugar syrup), 알코올성 음료 및 배출수 등 어떤 종류의 원수도 UV 기술로 효과적으로 처리된다.

한편, UV의 살균능력에 저항성이 있는 어떤 종류의 미생물도 아직 알려진바 없으며, 리스테리아(Listeria), 레지오넬라(Legionella) 및 크립토스포리듐(Cryptosporidium) 등의 병원성 미생물에 대해서도 충분한 살균 능력을 갖고 있다.

UV is the part of the electromagnetic spectrum between visible light and X-rays. The specific portion of the UV spectrum between 100-290nm (known as UV-C) has a strong germicidal effect, with peak effectiveness at 265nm. At these wavelengths UV eliminates microorganisms by penetrating their cell membranes and damaging the DNA, making them unable to reproduce and effectively killing them.
A typical UV disinfection system for process water or liquid ingredient consists of a UV lamp housed in a protective quartz sleeve and mounted within a cylindrical stainless steel chamber. The liquid to be treated enters at one end and passes along the entire length of the chamber before exiting at the other end. Virtually any liquid can be effectively treated with UV, including raw mains water, filtered process water, viscous sugar syrups, beverages and effluent.
There are no microorganisms known to be resistant to UV; this includes pathogenic bacteria such as Listeria, Legionella and Cryptosporidium(and its spores, which are resistant to chlorination).

UV 조사량(UV dose)

미생물의 활성화를 억제시키는데 필요한 최소한의 UV 조사량은 처리대상 물질별로 상이하며 일반적으로 mJ/㎠로 표시된다. 특정 미생물에 대한 UV 조사량은 과거의 오랜 실험 결과에 의하여 확립되어 있으며 UV 시스템의 형태 및 용량에 따라 결정된다. UV 시스템이 어떻게 구성되는가에 따라 결정되는 최적의 조사량은 다음 5가지 요인에 의존된다.

- 사용되는 UV의 에너지 출력(the energy output of the UV source)
- 챔버내의 물의 흐름량(the flow rate of the fluid through the treatment chamber)
- 처리되는 물의 이동거리(the transmission value of the fluid being treated)
- 처리용 챔버의 기하학적 배치(the geometry of the treatment chamber)
- 처리되는 물의 화학적 성분(the chemistry of the water)

이러한 조건들을 최적화함으로써 UV시스템은 대용량·소용량으로 크기를 결정, 점도성 있는 유체의 처리 유무, 용존성 고형물질 및 전분이나 당도를 함유한 물질의 적절한 처리에 적합하도록 각각의 사용자 환경을 고려하여 제작될 수 있다.

The UV irradiated dose necessary for microbial deactivation varies from one species to another and is measured in millijoules per square centimetre(mJ/㎠). Values for specific microorganisms have been experimentally established and are used to determine the type and size of UV system required.
The dose received by an organism in a UV treatment system is dependent on five main factors:
- The energy output of the UV source
- The flow rate of the fluid through the treatment chamber
- The transmission value(ability to transmit UV light) of the fluid being treated
- The geometry of the treatment chamber
- The chemistry of the water.
By optimising these criteria, a UV system can be tailored to effectively treat large or small flows, as well as viscous fluids, or those containing dissolved solids and high levels of starch or sugar compounds.

UV 기술의 원리(UV technologies) 

일반적으로 UV 램프에 관하여 2가지의 UV 기술(저압용 및 중간압력용)이 사용되며 저압용 램프는 ‘monochromatic’ UV output(254㎚의 단순파장으로 제한) 특성을 가지며 중간압력용 램프는 ‘polychromatic’ UV output(185∼400㎚ 사이에 분포) 특성을 갖는다.

UV 기술을 사용하면 잔존 발생물이 존재하지 않으므로 UV 처리  시스템의 공정상 설치되는 위치는 사용자 근접위치(point of use)가 가장 효과적이라 할 수 있다. 즉, 유입되는 미생물로부터 발생되는 오염물질은 철저히 파괴되므로 이후 처리 공정에서의 오염화 발생가능성은 극히 미미하다.

There are two main types of UV technology based on the UV lamps used: low pressure and medium pressure. Low pressure lamps have a ‘monochromatic’ UV output (limited to a single wavelength at 254㎚), whereas medium pressure lamps have a ‘polychromatic’ UV output (between 185∼400㎚).
As UV has no residual effect, the best position for a treatment system is immediately prior to the point of use. This ensures incoming microbiological contaminants are destroyed and there is a minimal chance of posttreatment contamination.

UV 기술의 적용 사례  

제약 및 음·식료 산업 분야에서의 UV 적용사례는 다음과 같다.

1) 물의 직접 접촉 분야(direct contact water)

도시용으로의 물 공급은 유해한 병원성 미생물이 포함되지 않아야 한다. 이외에 천연 스프링을 이용한 원수 채수 혹은 지하천공을 통한 개인용 수도장치의 사용은 일부 오염화되어 있다. 또한 식품 첨가제로 사용되거나 제품과 직접 접촉되어 사용된 물도 오염화가 진행될 수 있다.

이러한 물은 케미컬의 사용 없이도 UV 기술로 충분히 살균 및 소독되며, 공정상에서 이미 사용된 물도 별도 공정상에서 UV 처리 기술을 통하여 재이용수로 사용 가능하다. 즉, 비용 절약, 생산성 향상 등의 효과를 얻을 수 있다. 
  
Although municipal water supplies are normally free from harmful or pathogenic microorganisms, this should not be assumed. In addition, water from private sources such as natural springs or boreholes could also be contaminated.
Any water used as an ingredient, or that comes in direct contact with the product, can therefore be a source of contamination. UV disinfects this water without chemicals or pasteurisation. It also allows the re-use of process water, saving money and improving productivity without risking the quality of the product.

2) 세정용 린스(CIP rinse water)

외부의 물질을 세척하거나 살균용제로 사용되는 CIP(Clean-In-Place) 목적의 린스용 물은 미생물 관점에서 안전해야 한다. 일부 자동화처리 가능한 UV 살균시스템은 CIP 목적의 린스 공정과 연계되어 린스 처리된 물의 세척으로 인하여 미생물에 의한 오염이 발생되지 않는다.

It is essential that the CIP final rinse water used to flush out foreign matter and disinfecting solutions is microbiologically safe. Fully automated UV disinfection systems can be integrated with CIP rinse cycles to ensure final rinse water does not reintroduce microbiological contaminants.  

3) 필터장치의 살균·소독(Filter disinfection)

RO 멤브레인 및 활성탄탑(GAC)은 일반적으로 사용되는 중요한 여과 공정이나 박테리아 서식의 원천이 될 수 있다. 그러나 UV 기술은 RO 혹은 GAC 처리된 물의 살균 및 소독을 위한 효과적인 방법으로 오랫동안 사용되어 왔다.

GAC 필터는 간혹 공정수의 탈염 목적으로 사용되기도 하나 염소주입으로 인하여 발생되는 특유의 냄새를 제거할 필요가 있다. 탈염 목적의 GAC 필터 이전에 UV시스템을 설치하면 필터의 성능을 개선시키고 카본재료의 수명을 연장시켜 결국 운영비를 감소할 수 있다.

Reverse osmosis (RO) and granular activated carbon (GAC) are often used to filter process water, but can be a breeding ground for bacteria. UV is an effective way of disinfecting both stored RO and GAC filtered water and has been used in the process industries for many years.
GAC filters are also often used to dechlorinate process water, removing the ‘off’ flavours often associated with chlorine disinfection, meaning the flavour of the final product remains untainted and free from unwanted flavours or odours. Placing UV systems ahead of GAC filters used for dechlorination improves the performance of the filters and results in longer carbon runs, so decreasing operating costs. As the usual dose for removing free chlorine is 15 to 30 times higher than the normal disinfection dose, an important additional benefit of using UV dechlorination is a high level of UV disinfection, reduction in Total Organic Carbon (TOC) and improved overall water quality at point-of-use.

4) 총유기탄소량 절감(TOC reduction)

단파장(200㎚ 이하) UV의 사용은 물속에 함유되어 있는 낮은 분자량의 TOC 물질인 유기성분자 물질의 파괴에 매우 효과적이다. 이 과정은 2가지 방법으로 진행, 첫 번째는 direct photolysis에 의한 것으로 UV의 에너지에 의해 유기물질의 화학적 결합상태를 파괴시키는 방법이다. 두 번째는 물분자의 photolysis에 의한 것으로 물분자로부터 도전상태의 OH 수산화기를 분리·생성시켜 유기물질을 공격하는 방법이다.

Short UV wavelengths(below 200㎚) are highly effective at breaking down organic molecules present in water, commonly known as Total Organic Carbon (TOC), especially low molecular weight contaminants. This works in two ways: the first method is by direct photolysis, when energy from the UV actually breaks down chemical bonds within the organics; the second method is by the photolysis of water molecules, splitting them to create charged OH radicals, which also attack the organics.

5) 냉각수 살균 및 소독(Cooling media and chiller disinfection)

일부 정육과 낙농제품은 가열처리 및 음식 제조과정에서 오염화되기 쉬우며 이 경우 UV 기술은 냉각수와 접촉하게 하여 음식조리에서 오염화되는 것을 효과적으로 방지하는 방법으로 사용된다.

Some meat and dairy products are subject to contamination after heat treatment or cooking. UV provides an excellent way to protect foods from contamination by contactcooling fluids.

6) 설탕류 시럽(Sugar syrups)

설탕류 시럽도 미생물이 자랄 수 있는 원천이 된다. 실제로 당분 함량이 매우 높은 설탕류에서는 미생물 성장이 이루어지지 않으나 시럽자체가 희석이 된 후에는 잔존하고 있는 휴면상태의 포자가 활성화되기도 하여 미생물이 성장된다. 시럽과 희석수의 혼합수에 UV처리를 하면 휴면상태의 미생물 등이 활성화되는 것을 방지할 수 있다.

Sugar syrups can indeed be a prime breeding ground for microorganisms. Although syrups with very high sugar content do not support microbial growth, any dormant spores could become active after the syrup has been diluted. Treating the syrup and dilution water with UV prior to their use will ensure that any dormant microorganisms are properly deactivated.

7) 부드러운 음료(Liquid sweeteners)

수크로오스(sucrose) 재료의 당분체 역시 미생물이 자랄 수 있는 원천이 되나 UV 기술이 이러한 시럽을 처리하는데 사용된다.

Sucrose-based sweeteners can also be a prime breeding ground for microorganisms. UV systems are available specifically for treating these syrups.

8) De-aerated liquor 

De-aerated liquor는 양조제조의 일부공정에 주입되며 최종 패키지 공정에서도 사용된다. 이러한 액상의 물은 맥주원액에 직접 투입되어 gram negative bacteria에 의한 오염물질의 발생을 억제하는 기능을 한다.

De-aerated liquor is added as part of a high gravity brewing process, often in the packaging operation. This liquor is added directly to the beer so needs to be kept free from contamination by gram negative bacteria, which can cause off-flavors and acidity.

9) 맥주 효모의 준비(Yeast preparation)

맥주제조의 효모 준비과정에서 발생되는 문제는 hazes, altered fermentation, packaged beer에서의 surface membrane 등으로 알려져 있다. 효모(pitching yeast)에서의 Sacchoromyces라는 단세포는 쉽게 검출되는 haze의 원인이 될 수 있으며 UV기술로 이러한 다양한 현존하는 효모균 및 포자 등을 파괴할 수 있다
 
The problems associated with yeast preparation in breweries are well recognised and include hazes, altered fermentation and surface membranes on packaged beer. A single cell of Sacchoromyces(var. Turbidans) in 16 million cells of pitching yeast will cause detectable hazes. UV destroys all known yeasts and their spores.

10) 폐수처리(Waste Water)

제약 플랜트, 음·식료 제조 플랜트에서의 방류수는 환경에 유해한 케미컬의 사용 없이 처리 될 수 있다. 즉, 모든 배출수는 지역별 배출수 환경기준을 준수할 수 있다. 이미 언급한 바와 같이 UV 기술로 플랜트의 공정수는 배출목적으로 처리되거나 재이용처리 가능하다. 이를 통해 최종 발생되는 폐수량을 크게 절감한다.

Effluent from pharmaceutical, food and beverage facilities can be treated without the use of environmentally hazardous chemicals. This ensures all discharges meet with local environmental regulations. As already mentioned, because process water can be treated and re-used with UV, this also leads to a significant reduction in the amount of waste water produced.

결론(Conclusion) 

제약 산업, 음·식료 산업의 제조공정에서 점차 강화되고 있는 위생처리 기준을 준수하는 것은 각각의 플랜트에서 문제점으로 대두되고 있다. 플랜트 설비 및 시스템 설비에 대한 개선이 필요한 경우, 투자에 대한 신속한 투자회수의 가능성 및 제품자체 품질의 정량적인 개선 성과가 보장되어야 한다.

최종 제품에 대한 품질을 개선하고 제조 공정에서의 bio-security를 보장하려는 제조사의 UV 기술 사용은 경제성을 가지며 보다 현실적인 해결책이 된다. 먹는 물 공급의 살균 및 소독을 위해 충분히 검증된 기술이며 높은 순도의 물을 필요로 하는 플랜트 공정의 초순수 생산에 널리 적용되고 있다.

UV 기술은 설치하기가 용이하여 플랜트 운전에 최소한의 부담으로 작용하며 시스템의 정비보수 작업도 UV 램프의 교체만이 필요해 특별한 유지보수의 부담이 없다. 한편, 실제 운영도 수분 내 짧은 시간에 운전 가능할 정도로 간단한 운전으로, 일반 정비보수 인원만으로도 운전을 수행할 수 있다.

Meeting the increasingly rigorous hygiene standards required in the production of pharmaceutical, food and beverage products is a real challenge. If improvements need to be made to plant and equipment, they need to bring quick returns on the investment and measurable improvements in product quality.
For manufacturers seeking to improve the quality of the end product and to ensure the bio-security of their process, UV is an economic, realistic option. It is an established method of disinfecting drinking water, and is widely used for high purity applications where water of the highest quality is essential.
UV disinfection systems are easy to install, with minimum disruption to a plant. They need little maintenance; the only requirement being replacement of the UV lamps every 9∼12 months, depending on use. This is a simple operation that takes a few minutes and can be carried out by general maintenance staff.
